Microsatellites Associated to Gluten Protein Genes of Chilean Wheat Varieties: Description and Potential use as Bread Quality Markers

Abstract
Thirty-nine Chilean wheat genotypes were characterized by
analyzing the microsatellite alleles linked to a low molecular weight glutenin
(LMWG) gene and a g -gliadin pseudo-gene. Eight alleles for LMWG and nine for
g -gliadin loci were detected, including one null allele. Considering both
allele groups, a considerable proportion of the analyzed varieties (44%) showed
unique allelic combinations, suggesting this as an attractive fingerprinting method
for genetic identification purposes.
A portion of the genotypes characterized by SSR's was analyzed using
standard bread quality tests. These results were correlated with the alleles
of determined SSR patterns, using a methodology designed to compare qualitative
and quantitative variables. Statistically significant associations were found
between LMWG and sedimentation (0.51**), gluten-index (0.42**) and dry gluten
(0.35**) tests. In the case of g -gliadin alleles, statistically significant
correlations were detected with all the bread quality tests conducted (0.54**
for protein percentage, 0.47** for sedimentation, 0.52** for gluten-index and
0.49** for dry gluten).
As well, a statistically significant association (0.48**) was found between
LMWG and the "English Index" for bread quality, which is based on
high molecular weight glutenin (HMWG) patterns. On the other hand, there was
no association among LMWG and the "French Index", nor between g -gliadin and
the French and English indexes.

Secuencias de Microsatélites Asociadas a Genes de Proteínas de Reserva en Variedades Chilenas de Trigo Harinero: Descripción y Posible uso Como Marcadores de Calidad Panadera

Resumen
Se caracterizaron 39 genotipos chilenos de trigo
harinero mediante el análisis de alelos de microsatélites (SSR) asociados
a un gen de glutenina de bajo peso molecular (LMWG) y a un pseudo-gen de g -gliadina.
Se detectaron ocho alelos para el locus de LMWG y nueve para el de g -gliadina,
incluido un alelo nulo. Desde un punto de vista de la identificación genética,
un 44% de las variedades estudiadas presentaron combinaciones de alelos únicas.
Parte de los genotipos caracterizados por SSR fue sometido a pruebas tradicionales
de calidad panadera. Estos resultados fueron correlacionados con los alelos de
SSR determinados, usando una metodología que permite comparar variables cualitativas
con variables cuantitativas. Se encontraron asociaciones estadísticamente
significativas entre los alelos de LMWG con las pruebas de sedimentación
(0,51**), gluten index (0,42**) y gluten seco (0,35**). Para el caso de los alelos
de g -gliadinas se determinaron correlaciones estadísticamente significativas
con todas las pruebas de calidad realizadas (0,54** para porcentaje de proteína,
0,47** para sedimentación, 0,52** para gluten-index y 0,49** para gluten
seco). Así también, se encontró una correlación estadísticamente
significativa (0,48**) entre LMWG e índice inglés para calidad panadera,
que está basado en los patrones de gluteninas de alto peso molecular. Por
el contrario, no hubo correlación entre LMWG e índice francés,
ni entre g -gliadina y los índices francés e inglés.
