Genetic improvement of bread wheats ( Triticum aestivum L.) in the south central zone of Chile. IV. Characterization of varieties based on electrophoretic profiles of glutenins and gliadins

Abstract
Twenty-two wheat cultivars released between 1968 and 1996
by the Quilamapu Research Center of the Chilean Agriculture Research Institute,
were analyzed for their seed storage protein profiles, including the high
molecular weight glutenins (HMWG) and the acidic gliadins. HMWG showed a
good reproducibility compared to previous determinations, and a high diversity,
with 14 different protein bands in 12 different allelic combinations, corresponding
to the
loci
GluA1, GluB1 and GluD1. A correlation between HMWG alleles related to quality
and sedimentation values was observed, with the exception of some cultivars.
The electrophoretic screening of the 22 cultivars identified the a , b ,
d and w gliadin types. The similarity among some cultivars, in terms of
sharing HMWG or gliadin profiles, did not have any correlation with their
having the same progenitors or similar pedigrees. The combination of HMWG
and gliadin banding patterns represents a plausible method to differentiate
the majority of the cultivars included in this study.

MEJORAMIENTO DE TRIGOS HARINEROS ( Triticum aestivum L.) EN LA ZONA CENTRO SUR DE CHILE. IV. CARACTERIZACIÓN DE CULTIVARES DE TRIGOS BASADO EN PERFILES ELECTROFORÉTICOS DE GLUTENINAS Y GLIADINAS

Resumen
Se analizaron 22 cultivares de trigo liberados entre 1968
y 1996 en el Centro Regional de Investigación Quilamapu del Instituto
de Investigaciones Agropecuarias, desde el punto de vista de los perfiles
de proteínas de reserva de las semillas, incluyendo las gluteninas
de alto peso molecular (G-APM) y las gliadinas ácidas. Las G-APM mostraron
una buena reproducibilidad comparadas con determinaciones anteriores, y
una alta diversidad, con 14 bandas proteicas en 12 diferentes combinaciones
alélicas, correspondientes a los
loci
Glu-A1, Glu-B1 y Glu-D1. Se encontró correlación entre los alelos
relacionados a calidad y los valores de sedimentación, con excepción
de algunos cultivares. El análisis electroforético de los 22 cultivares
identificaron los tipos de gliadinas a , b , d y w . La similitud entre
algunos cultivares, en cuanto a presentar idénticos patrones de bandas
de G-APM o gliadinas, no tuvo un correlato en cuanto a tener iguales progenitores
o similar pedigree. La combinación de los patrones de G-APM y de gliadinas
representa un método aplicable para diferenciar la mayoría de
los cultivares considerados en este estudio.
